Tidepools were thus isolated from each other as well as the\nnearshore environment during the low tide period when experiments were\nconducted. Each experiment included 4 to 5 mussel removal (MR) tidepools\n(since 2002) and 4 to 5 mussel control (MC) tidepools with natural mussel\ndensities. \\u00a0In June 2010, we performed daytime NH4+ tracer experiments\nand in August 2010 nighttime experiments using the same tidepools. The\nfollowing year (July 2011) these experiments were repeated with the addition\nof bottle incubations (see below) to evaluate the effects of suspended\ntidepool components and extended sampling for 6 days after the initial 15N\naddition to test for long-term retention of NH4+. \\u00a0During the 2011\nexperiments, unforeseen rain reduced the salinity in some pools by up to 51%,\nand we have attempted to correct for the expected dilution of NH4+ in our\ntidepool rate calculations.\n \nBecause isotope enrichment levels were relatively low, we used the\nconventional delta notation instead of atom% to describe variations in 15N\nenrichment (where delta-15NH4+ = {(15N:14N sample \\u00f7 15N:14N standard)\n-1}\\u00d71000\\u2030, where the standard is atmospheric N2. \\u00a0Tracer\nlabeled ammonium chloride (15NH4Cl) was added to the pools to approximate a\n1000\\u2030 enrichment in 2010 (doubling the 15N-NH4+ concentration) and a\n2000\\u2030 enrichment in 2011(tripling the 15N-NH4+ concentration). 15N\nnatural abundance is only 0.365% and these tracer additions thus had a\nnegligible effect on the overall NH4+ concentrations increasing them by only\n~0.4% and ~0.8%, respectively. Tidepool volumes were estimated\nspectrophotometrically using varying concentrations of food dye (Pfister\n1995). Together with estimates of NH4+ concentration (from 2009 data), we\nestimated the tracer addition required to achieve the targeted 15N\nenrichments. However, the actual initial enrichments varied substantially,\n684.4 - 2406.4\\u2030 in 2010 and 781.4 - 3880.2\\u2030 in 2011, likely due to\nerror in tidepool volume estimation and natural variations in initial NH4+\nconcentrations. Fortunately, we sampled immediately following each tracer\naddition allowing for the determination of the true initial 15N enrichment.\n\\u00a0\n \nPrior to tracer addition at ebb tide, 100 mL of tidepool water was syringe-\nfiltered (Whatman GF/F) into separate HDPE bottles for natural abundance\n15NH4+ and concentration determination. \\u00a0To each pool, tracer 15NH4+ was\nthen added and distributed by stirring with a stick. Water samples were\nimmediately collected for measuring initial 15N enrichment and subsequently at\n2, 4, and 6 hour intervals to determine isotope and concentration time\ncourses. All water samples were frozen until analysis. Tidepool oxygen, pH,\nand temperature (Hach HQ4D) were also collected at ~ 2 h intervals throughout\nthe experiment.\n \nIn 2011 we also assessed the contribution of the suspended microbial community\nto NH4+ cycling by enclosing tidepool water in a 250 mL transparent\npolycarbonate incubation bottle. Following tracer addition, the bottle was\nfilled, then left to float in the tidepool for the duration of the experiment.\nSamples from bottles were filtered as described both immediately after\ncontainment and at the end of the experiment (~6 h later).\n \nWe assessed macroalgal contribution to NH4+ removal by transplanting two\ntidepool-dwelling algae species. \\u00a0Prionitis sternbergii were sampled 2\nweeks prior to the experiment for baseline natural abundance 15N values and\ntransplanted into the pools with Z-Spar Epoxy (Pfister 2007). On the day of\nthe experiment, the red-alga, Corallina vancouveriensis from a single source\npatch, was also sampled for 15N natural abundance, inserted into pieces of\nStyrofoam, and floated in each pool.\n \nAt the end of each experiment (~6 h sampling point), we sampled tidepool\nparticulate organic material (POM) by filtering through combusted GF/F filters\nuntil they clogged (~ 600 mL), comparing these samples with POM similarly\nsampled from the immediate nearshore. \\u00a0Floating Corallina spp. samples\nwere collected into clean Eppendorf tubes, and similar sized pieces of\nPrionitis spp. were collected from each pool into clean foil packets.\\u00a0  \n We evaluated the extent of longer-term 15N tracer retention in 2011 by\nsampling tidepool water, POM and transplanted Prionitis 1, 3, and 6 days\nfollowing tracer addition. Tidepools were thus isolated from each other as well as the\nnearshore environment during the low tide period when experiments were\nconducted. Each experiment included 4 to 5 mussel removal (MR) tidepools\n(since 2002) and 4 to 5 mussel control (MC) tidepools with natural mussel\ndensities. \\u00a0In June 2010, we performed daytime NH4+ tracer experiments\nand in August 2010 nighttime experiments using the same tidepools. The\nfollowing year (July 2011) these experiments were repeated with the addition\nof bottle incubations (see below) to evaluate the effects of suspended\ntidepool components and extended sampling for 6 days after the initial 15N\naddition to test for long-term retention of NH4+. \\u00a0During the 2011\nexperiments, unforeseen rain reduced the salinity in some pools by up to 51%,\nand we have attempted to correct for the expected dilution of NH4+ in our\ntidepool rate calculations.\n \nBecause isotope enrichment levels were relatively low, we used the\nconventional delta notation instead of atom% to describe variations in 15N\nenrichment (where delta-15NH4+ = {(15N:14N sample \\u00f7 15N:14N standard)\n-1}\\u00d71000\\u2030, where the standard is atmospheric N2. \\u00a0Tracer\nlabeled ammonium chloride (15NH4Cl) was added to the pools to approximate a\n1000\\u2030 enrichment in 2010 (doubling the 15N-NH4+ concentration) and a\n2000\\u2030 enrichment in 2011(tripling the 15N-NH4+ concentration). 15N\nnatural abundance is only 0.365% and these tracer additions thus had a\nnegligible effect on the overall NH4+ concentrations increasing them by only\n~0.4% and ~0.8%, respectively. Tidepool volumes were estimated\nspectrophotometrically using varying concentrations of food dye (Pfister\n1995). Together with estimates of NH4+ concentration (from 2009 data), we\nestimated the tracer addition required to achieve the targeted 15N\nenrichments. However, the actual initial enrichments varied substantially,\n684.4 - 2406.4\\u2030 in 2010 and 781.4 - 3880.2\\u2030 in 2011, likely due to\nerror in tidepool volume estimation and natural variations in initial NH4+\nconcentrations. Fortunately, we sampled immediately following each tracer\naddition allowing for the determination of the true initial 15N enrichment.\n\\u00a0\n \nPrior to tracer addition at ebb tide, 100 mL of tidepool water was syringe-\nfiltered (Whatman GF/F) into separate HDPE bottles for natural abundance\n15NH4+ and concentration determination. \\u00a0To each pool, tracer 15NH4+ was\nthen added and distributed by stirring with a stick. Water samples were\nimmediately collected for measuring initial 15N enrichment and subsequently at\n2, 4, and 6 hour intervals to determine isotope and concentration time\ncourses. All water samples were frozen until analysis. Tidepool oxygen, pH,\nand temperature (Hach HQ4D) were also collected at ~ 2 h intervals throughout\nthe experiment.\n \nIn 2011 we also assessed the contribution of the suspended microbial community\nto NH4+ cycling by enclosing tidepool water in a 250 mL transparent\npolycarbonate incubation bottle. Following tracer addition, the bottle was\nfilled, then left to float in the tidepool for the duration of the experiment.\nSamples from bottles were filtered as described both immediately after\ncontainment and at the end of the experiment (~6 h later).\n \nWe assessed macroalgal contribution to NH4+ removal by transplanting two\ntidepool-dwelling algae species. \\u00a0Prionitis sternbergii were sampled 2\nweeks prior to the experiment for baseline natural abundance 15N values and\ntransplanted into the pools with Z-Spar Epoxy (Pfister 2007). On the day of\nthe experiment, the red-alga, Corallina vancouveriensis from a single source\npatch, was also sampled for 15N natural abundance, inserted into pieces of\nStyrofoam, and floated in each pool.\n \nAt the end of each experiment (~6 h sampling point), we sampled tidepool\nparticulate organic material (POM) by filtering through combusted GF/F filters\nuntil they clogged (~ 600 mL), comparing these samples with POM similarly\nsampled from the immediate nearshore. \\u00a0Floating Corallina spp. samples\nwere collected into clean Eppendorf tubes, and similar sized pieces of\nPrionitis spp. were collected from each pool into clean foil packets.\\u00a0  \n We evaluated the extent of longer-term 15N tracer retention in 2011 by\nsampling tidepool water, POM and transplanted Prionitis 1, 3, and 6 days\nfollowing tracer addition. Tidepools were thus isolated from each other as well as the\nnearshore environment during the low tide period when experiments were\nconducted. Each experiment included 4 to 5 mussel removal (MR) tidepools\n(since 2002) and 4 to 5 mussel control (MC) tidepools with natural mussel\ndensities. \\u00a0In June 2010, we performed daytime NH4+ tracer experiments\nand in August 2010 nighttime experiments using the same tidepools. The\nfollowing year (July 2011) these experiments were repeated with the addition\nof bottle incubations (see below) to evaluate the effects of suspended\ntidepool components and extended sampling for 6 days after the initial 15N\naddition to test for long-term retention of NH4+. \\u00a0During the 2011\nexperiments, unforeseen rain reduced the salinity in some pools by up to 51%,\nand we have attempted to correct for the expected dilution of NH4+ in our\ntidepool rate calculations.\n \nBecause isotope enrichment levels were relatively low, we used the\nconventional delta notation instead of atom% to describe variations in 15N\nenrichment (where delta-15NH4+ = {(15N:14N sample \\u00f7 15N:14N standard)\n-1}\\u00d71000\\u2030, where the standard is atmospheric N2. \\u00a0Tracer\nlabeled ammonium chloride (15NH4Cl) was added to the pools to approximate a\n1000\\u2030 enrichment in 2010 (doubling the 15N-NH4+ concentration) and a\n2000\\u2030 enrichment in 2011(tripling the 15N-NH4+ concentration). 15N\nnatural abundance is only 0.365% and these tracer additions thus had a\nnegligible effect on the overall NH4+ concentrations increasing them by only\n~0.4% and ~0.8%, respectively. Tidepool volumes were estimated\nspectrophotometrically using varying concentrations of food dye (Pfister\n1995). Together with estimates of NH4+ concentration (from 2009 data), we\nestimated the tracer addition required to achieve the targeted 15N\nenrichments. However, the actual initial enrichments varied substantially,\n684.4 - 2406.4\\u2030 in 2010 and 781.4 - 3880.2\\u2030 in 2011, likely due to\nerror in tidepool volume estimation and natural variations in initial NH4+\nconcentrations. Fortunately, we sampled immediately following each tracer\naddition allowing for the determination of the true initial 15N enrichment.\n\\u00a0\n \nPrior to tracer addition at ebb tide, 100 mL of tidepool water was syringe-\nfiltered (Whatman GF/F) into separate HDPE bottles for natural abundance\n15NH4+ and concentration determination. \\u00a0To each pool, tracer 15NH4+ was\nthen added and distributed by stirring with a stick. Water samples were\nimmediately collected for measuring initial 15N enrichment and subsequently at\n2, 4, and 6 hour intervals to determine isotope and concentration time\ncourses. All water samples were frozen until analysis. Tidepool oxygen, pH,\nand temperature (Hach HQ4D) were also collected at ~ 2 h intervals throughout\nthe experiment.\n \nIn 2011 we also assessed the contribution of the suspended microbial community\nto NH4+ cycling by enclosing tidepool water in a 250 mL transparent\npolycarbonate incubation bottle. Following tracer addition, the bottle was\nfilled, then left to float in the tidepool for the duration of the experiment.\nSamples from bottles were filtered as described both immediately after\ncontainment and at the end of the experiment (~6 h later).\n \nWe assessed macroalgal contribution to NH4+ removal by transplanting two\ntidepool-dwelling algae species. \\u00a0Prionitis sternbergii were sampled 2\nweeks prior to the experiment for baseline natural abundance 15N values and\ntransplanted into the pools with Z-Spar Epoxy (Pfister 2007). On the day of\nthe experiment, the red-alga, Corallina vancouveriensis from a single source\npatch, was also sampled for 15N natural abundance, inserted into pieces of\nStyrofoam, and floated in each pool.\n \nAt the end of each experiment (~6 h sampling point), we sampled tidepool\nparticulate organic material (POM) by filtering through combusted GF/F filters\nuntil they clogged (~ 600 mL), comparing these samples with POM similarly\nsampled from the immediate nearshore. \\u00a0Floating Corallina spp. samples\nwere collected into clean Eppendorf tubes, and similar sized pieces of\nPrionitis spp. were collected from each pool into clean foil packets.\\u00a0  \n We evaluated the extent of longer-term 15N tracer retention in 2011 by\nsampling tidepool water, POM and transplanted Prionitis 1, 3, and 6 days\nfollowing tracer addition. To enhance the activity of nitrate-\nrespiring anaerobic bacteria, an NO3- (5 mm) and H2(aq) (1.30 mM)-enriched\nmedium was introduced in the high-pressure incubations under strictly\nanaerobic conditions. Dissolved HCO3- (7.3 mm, 13C labeled) was used as added\ncarbon source. Vent fluids were introduced at a flow rate of 0.042 mL/min,\nwhile growth medium was added at a rate of 0.0042 mL/min. The two sets of\nexperiments were performed for 356 (Crab Spa) and 50 hours (Alvinella). Here we quantify rates of N2 fixation as well as its contribution to export production along a zonal transect in the Western Tropical South Pacific (WTSP) Ocean using N isotope (\\u201cd15N\\u201d) budgets. Comparing measurements of water column nitrate+nitrite d15N with the d15N of sinking particulate N at a western, central, and eastern station, these d15N budgets indicate high, modest, and low rates of N2 fixation at the respective stations. The results also imply that N2 fixation supports exceptionally high, i.e., >50%, of export production at the western and central stations, which are also proximal to the largest iron sources.
