This study was conducted to supplement the gene-based and metagenomic studies. However, the results from sequenced reverse transcribe 16S rRNA demonstrate the sensitivity of extraction and amplification to external nucleic acids and attest to the challenges in working with low biomass environmental samples. The bulk media was prepared with the same components and a portion was treated with Chelex-100 to remove metals, while the other portion remained un-chelexed. Half of each treatment was spiked with labeled \\u2075\\u2077Fe and either 0.2 \\u03bcm or 0.02 \\u03bcm filtered for comparison of the dissolved and soluble fractions. Microbiology sampling were conducted during special CARIACO cruises (distinct from the monthly, core sampling, time-series cruises) from November 1995 to November 2015. The specialized microbiology cruises were usually during May and November, and could be opportunistic or process-driven. CTD\\u2019s Salinity and Oxygen where calibrated with in-situ measurements (see Acquisition Description). This dataset is complimentary to the monthly \\u201cCTD Composite Profiles\\u201d (https://www.bco-dmo.org/dataset/3092), and many fields are very similar to that data-base. (EAGER: Iron-Virus Interactions in the Ocean) This data was collected as part of a study investigating the source of iron to bacteriophage (phage for short, or viruses that infect and kill bacteria) progeny. Evidence from a phage that infects E. coli shows iron incorporated into the tail fiber structure. This study aims at identifying whether the source of the iron is environmental or bacterially derived. E. coli bacterial cultures were grown in minimal media spiked with 10 \\u00b5M 57FeSO4 then infected with phage T4 or T5. Sea ice cores were drilled with a MARK II ice auger (Kovacs Enterprise). (1995) and Heller et al. (2017).\\r\\n\\r\\nFrom published and ongoing work in the Peru and Chile oxygen minimum zones an optimized methodology for Fe(II) detection by luminol has been described in Croot et al. (2019). This work details some of the challenges and limitations of using luminol for dFe(II) detection. It should be noted that as Fe(II) is a transient redox species with a short half life in oxygenated seawater that is dependent on dissolved oxygen and pH, it is not possible to archive (preserve) samples for later analysis. (Biopolymers as carrier phases for selected natural radionuclides (of Th, Pa, Pb, Po, Be) in diatoms and coccolithophores) To study the binding mechanisms of radionuclides to organic moieties in colloidal organic matter (COM),marine colloids (1 kDa\\u20130.2 \\u03bcm) were isolated by cross-flow ultrafiltration from seawater of the west Pacific Ocean and the northern Gulf of Mexico. For the same purpose, exopolymeric substances (EPS) produced by laboratory cultured diatoms were collected as well. In our study areas, colloidal organic carbon (COC) concentrations ranged from 6.5 to 202 \\u03bcg-C/L in the Pacific Ocean, and were 808 \\u03bcg-C/L in the Gulf of Mexico. The COM compositions (organic carbon, organic nitrogen, proteins, total hydrolysable amino acids, total polysaccharides, uronic acids, hydroxamate siderophores, hydroquinone) were quantified to examine the relationships between partition coefficients (Kc) of five different radionuclides, 234Th, 233Pa, 210Pb, 210Po and 7Be, and concentration ratios to COC of individual chelating biomolecules that could potentially act as a chelating moiety. The range of partition coefficients (Kc, reported as logKc) of radionuclides between water and the different colloidal materials was 5.12 to 5.85 for 234Th, 5.19 to 6.01 for 233Pa, 4.21 to 4.85 for 210Pb, 4.87 to 5.68 for 210Po, and 4.49 to 4.92 for 7Be, similar to values previously reported for lab and field determinations under different particle concentrations. While any relationship obtained between Kc and abundance of specific moieties could not be taken as proving the existence of colloidal organic binding ligands for the different radionuclides, it could suggest possible organic moieties involved in the scavenging of these natural radionuclides. Together with results from isoelectric focusing of radiolabeled COM, we conclude that binding to different biomolecules is nuclide-specific, with colloidal hydroxamate siderophoric moieties being important for the binding of Th and Pa radionuclides. Hydroquinones/ quinone (HQ/Q) facilitated redox and chelation reactions seem to be involved in the binding of Pa and Be. However, the actual mechanisms are not clear. Individual amino acids, proteins, total polysaccharides and uronic acids did not yield significant relationships with logKc values of the different radionuclides.
