Samples were collected to characterize vertical variations in archaeal lipid biomarkers and evaluate the relationship between TEX86 paleotemperature estimates and ambient water column temperature. In situ temperatures were obtained from CTD measurements, TEX86 values were calculated from the relative abundances of GDGT-1, GDGT-2, GDGT-3, and the crenarchaeol regioisomer, and TEX86-derived temperatures were reconstructed using a published calibration equation. These data were generated to assess how environmental gradients influence archaeal lipid distributions and the accuracy of TEX86 temperature reconstructions in the upper ocean.\n\ncdm_data_type = Other\nVARIABLES:\ntime (Datetime_of_sampling, seconds since 1970-01-01T00:00:00Z)\nlatitude (degrees_north)\nlongitude (degrees_east)\nStation_ID (unitless)\ndepth (Depth_m, m)\ninsitu_T_degC (degrees Celsius (°C))\nTEX86_derived_temperature_degC (degrees Celsius (°C))\nIsoprenoid_ng_per_L (nano gram per liter (ng/L))\n | https://erddap.bco-dmo.org/erddap/metadata/fgdc/xml/bcodmo_dataset_997800_v1_fgdc.xml | https://erddap.bco-dmo.org/erddap/metadata/iso19115/xml/bcodmo_dataset_997800_v1_iso19115.xml | https://erddap.bco-dmo.org/erddap/info/bcodmo_dataset_997800_v1/index.htmlTable | https://osprey.bco-dmo.org/dataset/997800

However, in situ measurements of N2O concentrations in the northeastern Pacific Ocean are scarce, and the processes controlling its production and consumption remain poorly constrained. During the R/V Kilo Moana cruise KM1314 (August 2013), vertical profiles were collected at Stations 1, 5, 8, and 13 from the surface to 300 m depth. The dataset includes dissolved N2O concentrations and isotopic measurements, including site preference and related isotope parameters. These observations provide constraints on the pathways and dynamics of N2O cycling in the upper water column and support investigation of nitrification and other microbial processes regulating marine N2O production in this region.\n\ncdm_data_type = Other\nVARIABLES:\ntime (Datetime_of_sampling, seconds since 1970-01-01T00:00:00Z)\nlatitude (degrees_north)\nlongitude (degrees_east)\nStation_ID (unitless)\ndepth (m)\nN2O_Concentration (nmol per milliliter (nmol/L))\nd15Na (permille (‰))\nd15Nb (permille (‰))\nSp (permille (‰))\nd15Nbulk (permille (‰))\nd17O (permille (‰))\nd18O (permille (‰))\n | https://erddap.bco-dmo.org/erddap/metadata/fgdc/xml/bcodmo_dataset_997809_v1_fgdc.xml | https://erddap.bco-dmo.org/erddap/metadata/iso19115/xml/bcodmo_dataset_997809_v1_iso19115.xml | https://erddap.bco-dmo.org/erddap/info/bcodmo_dataset_997809_v1/index.htmlTable | https://osprey.bco-dmo.org/dataset/997809

One liter of seawater from each depth was filtered onto 0.22 μm Sterivex filters and stored at −80 °C prior to DNA extraction using a modified manual extraction protocol. Archaeal and bacterial amoA gene abundances were quantified by quantitative PCR using primer sets CrenAmoAQ-F/CrenAmoAModR and AmoA1-F/AmoA2-R on a LightCycler platform. Standard curves, amplification procedures, melting curve analyses, and gel electrophoresis verification followed established protocols. These measurements characterize vertical distributions of ammonia-oxidizing microorganisms in the upper 300 m of the water column and support analyses of nitrification activity and microbial community structure in the oligotrophic North Pacific.\n\ncdm_data_type = Other\nVARIABLES:\ntime (Datetime_of_sampling, seconds since 1970-01-01T00:00:00Z)\nlatitude (degrees_north)\nlongitude (degrees_east)\nStation_ID (unitless)\ndepth (Depth_m, m)\nAOA_amoA_Calculated_Abundance (copies per milliliter (copies/mL))\nstdev_AOA (copies per milliliter (copies/mL))\nAOB_amoA_Calculated_Abundance (copies per milliliter (copies/mL))\nstdev_AOB (copies per milliliter (copies/mL))\n | https://erddap.bco-dmo.org/erddap/metadata/fgdc/xml/bcodmo_dataset_997806_v1_fgdc.xml | https://erddap.bco-dmo.org/erddap/metadata/iso19115/xml/bcodmo_dataset_997806_v1_iso19115.xml | https://erddap.bco-dmo.org/erddap/info/bcodmo_dataset_997806_v1/index.htmlTable | https://osprey.bco-dmo.org/dataset/997806

| log in | [Depth profiles of total prokaryotic cell counts KM1314] - Depth profiles of total prokaryotic cell counts from R/V Kilo Moana KM1314 in the North Pacific Ocean (Seattle to Honolulu, Line P, Station ALOHA), Aug-Sept 2013 (Significance of nitrification in shaping planktonic biodiversity in the ocean) | This dataset reports depth-resolved total prokaryotic cell counts collected at Stations 5, 8, and 13 in the northeastern Pacific Ocean during the R/V Kilo Moana cruise KM1314 in August 2013. Seawater samples (2 mL) from multiple depths were preserved with glutaraldehyde (2% final concentration), filtered onto 0.02 μm Anodisc filters, and stained with a Moviol–SYBR Green mixture. Cells were enumerated using epifluorescence microscopy by counting at least 20 randomly selected fields per sample (10–100 cells per field). The data characterize vertical variations in prokaryotic abundance in the upper 300 m of the water column in the oligotrophic North Pacific.\n\ncdm_data_type = Other\nVARIABLES:\ntime (Datetime_of_sampling, seconds since 1970-01-01T00:00:00Z)\nlatitude (degrees_north)\nlongitude (degrees_east)\nStation_ID (unitless)\ndepth (m)\nCell_counts (10^5 cells per milliliter (10^5 cells/mL))\nCell_counts_stdev (10^5 cells per milliliter (10^5 cells/mL))\n | BCO-DMO | bcodmo_dataset_997803_v1 |
