The bulk media was prepared with the same components and a portion was treated with Chelex-100 to remove metals, while the other portion remained un-chelexed. Half of each treatment was spiked with labeled \\u2075\\u2077Fe and either 0.2 \\u03bcm or 0.02 \\u03bcm filtered for comparison of the dissolved and soluble fractions. This dataset\\u00a0consists of mitochondrial sequence data and specimen\ninformation for two species of copepods, Haloptilus longicornis and\nPleuromamma xiphias, collected at an open ocean time series site in the North\nPacific Subtropical Gyre (station ALOHA, 22.45N, 158W) during 11 of the\nroutine Hawai'i Ocean Time-series (HOT) research cruises from September of\n2012 to October of 2013 (HOT-246 to HOT-256).\\u00a0Data for Haloptilus\nlongicornis include\\u00a0a 546 base-pair fragment of mitochondrial cytochrome\nc oxidase subunit II for each of 483 individuals (mean of 44 animals per\ncruise). Data for Pleuromamma xiphias include\\u00a0a 551 base-pair fragment of\nmitochondrial cytochrome c oxidase subunit I for each of 510 individuals (mean\nof 46 animals per cruise). Information is also provided on the HOT cruise\nnumber, date, and specific tow from which each individual was collected. This data was collected as part of a study investigating the source of iron to bacteriophage (phage for short, or viruses that infect and kill bacteria) progeny. Evidence from a phage that infects E. coli shows iron incorporated into the tail fiber structure. This study aims at identifying whether the source of the iron is environmental or bacterially derived. E. coli bacterial cultures were grown in minimal media spiked with 10 \\u00b5M 57FeSO4 then infected with phage T4 or T5. The phages were purified by methods of centrifugation, filtration, density-dependent ultracentrifugation, and dialyzing. Fluids were collected using isobaric\ngas-tight samplers [Seewald et al., 2002] prior to their transfer to the\nshipboard continuous culture system [Foustoukos and Perez-Rodriguez, 2015].\nHere, high-pressure incubations (250 bars) were conducted at mesophilic (30\n\\u00baC) and thermophilic (50 \\u00baC) conditions to constrain the function\nand metabolic rates of denitrifying and DNRA microbial communities residing at\nCrab Spa and Alvinella, respectively. To enhance the activity of nitrate-\nrespiring anaerobic bacteria, an NO3- (5 mm) and H2(aq) (1.30 mM)-enriched\nmedium was introduced in the high-pressure incubations under strictly\nanaerobic conditions. Dissolved HCO3- (7.3 mm, 13C labeled) was used as added\ncarbon source. Vent fluids were introduced at a flow rate of 0.042 mL/min,\nwhile growth medium was added at a rate of 0.0042 mL/min. The two sets of\nexperiments were performed for 356 (Crab Spa) and 50 hours (Alvinella). Direct\ncell counts were conducted by staining cells with 0.1% acridine orange and\ncounting them with a fluorescence microscope. 15N/14N isotopic analysis of the\nNO3-, NH4+ and biomass were conducted with a Thermo Scientific Delta VPlus\nmass spectrometer and CE Instruments NA 2500 series elemental analyzer (EA).\n \nReferences:\n \nFoustoukos, D., and I.
